NAD+ NADH Assay

This colorimetric assay detects NAD+ and NADH from peripheral or capillary whole blood. Sample input is 150ul of whole blood stored in heparin or EDTA. Assay is compatible with most self-collection lancets. Using an enzymatic reaction, NAD+ and NADH are measured at 570-573nm and compared to a standard curve to derive concentration. Assay requires approximately 20 minutes hands-on time, with a total time of approximately 3 hours.  Extracted samples may be stored for 7 days (4-8C) or 4 weeks (-20C). Standards, positive control, extraction buffer, stabilizers, reference material, and reagents included.

  • Instruments:
    • Spectrophotometer capable of measuring 570-573nm wavelength. Plate spectrophotometer preferred. 5-10 flashes per measurement.
    • Microcentrifuge with cooling to 4C (20,000 x g)
  • Standard Curve: 5 data points, in duplicate
  • Samples: Perform in duplicate

Note: 1) Final steps of assay must be performed in dim-light as reagents are photo-sensitive.  2) Do not use molecular biology grade sterile tubes or “lobind” tubes.
